Can I calculate the shipping cost according to the total weight of the order?
 
For example:
 
Items weighing up to 250g £1.62
Items weighing up to 500g £2.14
Items weighing up to 5kg £6.95

In the most simplest term, the answer would be YES. BUT, several other factors would come into play.
 
1. Type of Courier service being used.
1A Number of courier service you'll be using.
2. Size of the container or box.
3. Air or land delivery.
4. Will you be offering coupon FREE shipping based on amount of purchase.
 
Many of these questions can be answered by reviewing various courier service, UPS and etc.. One thing for sure SHIPPING configuration will be the most challenging aspect you will encounter to function properly.

Shipping Methods
 
In the weight section of the shipping method you would specify min 0 max 250 and show cost for the first item
min 250.001max 500 for the next with the cost
and min 500.001 max 5000 plus cost for the last
 
Hope this gets you started - a little exploration and you will find much other choice as Tony mentions.

Where is this weight-section?
When I go to the shipping modules and I edit my shipping type, I only see things like 'General', 'Users Types' and 'Sites'.

You need Shipping METHODS - found under the 'products' menu. Click Shipping Settings.
It is a bit confusing.
 
It will show breadcrumbs as:
Administration > Shipping Modules
 
- clicking further you can get to [ Administration > Shipping Modules > United Kingdom > Shipping Types ] where United Kingdom is one of my Shipping Ruls ( Administration > Static Tables > Shipping Rules > Edit Shipping Rule) which you must have set up to start.
 
Have you read the manual about setting up shipping? Recommend you do if not

Yeah, I have read the manual, but couldn't find what I was looking for.
 
I found shipping methods under the products menu, that was from where I was working along all the time. I don't have United Kingdom as shipping module (it's not a shipping rule), but I do have 'Custom shipping':

From there if I click on 'Shipping methods' I get to 'Shipping types':

And I don't get it what you mean with 'where United Kingdom' is one of my shipping rules. the only think I can setup for a shipping rule is a name and to which countries it applies:

The only place where I can do something with my shipping rules is at the 'shipping restrictions' at the 'stock & shipping'-tab when I edit a product:
